The beginning of the war found the regiment near Chojnice and Tuchola. As a result of heavy fights with Guderian's armored units in Bory Tucholskie (Bukowiec, Górna Grupa, Poledno) and air raids, on September 7, 1939, the regiment ceased to exist as a compact unit (losses reached 40% of the manpower). Despite this, 150 lancers of the 16th regiment fought at Kock.
The Reserve Center mobilized 2 horse squadrons, 3 fast ones and two platoons of cyclists. These units fought in the ranks of the "Garwolin" Cavalry Group and were disbanded after the Second Battle of Tomaszów Lubelski.
45 Crosses of Valor were awarded for combat operations.
